What Is RNA Sequencing?
RNA sequencing (RNA-seq) involves the sequencing of RNA transcripts in a given sample which facilitates their quantification in downstream analyses.
RNA comes in many forms, but the primary form for transcriptomic analysis is messenger RNA (mRNA). mRNA is transcribed from genes in DNA and contains the information to make a protein. Thus, identifying and quantifying mRNA transcripts in a sample gives us a picture of what genes are activated in that sample.
Bulk RNA-seq using Illumina’s NovaSeq X Series and NovaSeq 6000 Series platforms offered by Sampled allow for sequencing of all RNA species (mRNA and non-coding RNAs), giving not only a view of gene activation but also a picture of the regulatory RNA landscape within a sample.
Bulk RNA-seq can be performed on a sample containing many cells, but with the power of the 10x Genomics transcriptomics Chromium X and Chromium Connect platforms, Sampled also provides single-cell RNA sequencing (scRNA-seq); that’s a snapshot of gene expression at single-cell resolution. How Does RNA Sequencing Work?
Sample Preparation
Sampled works with a wide variety of sample types, from clinical samples to samples from academic research labs. The starting material is RNA, which can be extracted in-house or received directly from clients. Extracted RNA must pass quality control analysis before being taken forward for further processing.
Library Preparation
High-quality RNA is fragmented into smaller, more manageable sizes before end repair and polyadenylation are performed. Then the RNA is converted to complementary DNA through a process called reverse transcription before being barcoded with DNA adapters (small, artificial DNA sequences).
Sequencing
Given the variety in outputs for RNA-seq, such as scRNA-seq, Sampled’s team of experts works with the client to decide what platform is best for their needs, as well as offering advice on depth and coverage tailored to the specific goal of the experiment. What Are the Input Requirements for RNA Sequencing?
