Extraction
Sampled’s extraction services encompass the retrieval of high-quality RNA and DNA from diverse sample types. These nucleic acids serve as the starting point for various downstream analyses, ensuring optimal results. Stringent quality assurance measures are employed to guarantee top-quality data.
We accept samples for extraction at either room temperature or frozen, depending on the sample type, offering flexibility for immediate processing or storage. The processing method depends on the sample type and client’s timeline, with options for automated and manual extraction. Rigorous quality control, including functional DNA quality checks, ensures suitability for downstream analyses. After quality control, our team can proceed with analysis, storage, or shipping, with our logistics team handling shipping logistics to enable clients to concentrate on their projects.
Fractionation
Fractionation involves separating whole blood into distinct fractions, often acquiring serum or plasma. This process employs centrifugation to isolate plasma and serum from the rest of the sample. Sampled efficiently conducts large-scale fractionation, ensuring a high level of precision and swift turnaround times.
All samples for fractionation should arrive at room temperature to prevent hemolysis during thawing. Upon receipt, whole blood samples are kept at room temperature and processed within 48 hours. Fractionation is conducted using our automated process, led by our technical experts and the Hamilton easyBlood STARlet™ platform, ensuring precise and safe aliquoting of serum or plasma by accurately recognizing the different layers of fractionated blood. Rapid and secure sample processing is guaranteed through our technical proficiency and advanced instrumentation. After fractionation, samples can be directed for analysis, storage, or shipment to the client or a third party.
Normalization
Normalization is a critical step that often follows nucleotide extraction and involves adjusting sample volume and concentration to meet specific application requirements. Sampled’s experts can tailor normalization to clients’ needs or optimize it for in-house analyses. Proper normalization is vital to avoid downstream analysis failure, reagent wastage, and time loss, making Sampled a trusted choice for precise sample handling.
We accommodate samples extracted in-house or pre-extracted by our clients for normalization. Regardless of the source, we maintain proper temperature conditions during sample handling, with each sample undergoing QC for concentration and integrity. The parameters for normalization are tailored to factors including sample concentration/volume, downstream platform requirements, and client specifications. Our expert team employs high-end automated instrumentation to ensure efficient and precise normalization. QC checks, including concentration and quality assessments, are routinely performed on DNA and RNA. Following normalization and quality control, samples can be efficiently directed for aliquoting, storage, shipping, or laboratory analysis. Our dedicated logistics team skillfully manages the shipping process, enabling clients to focus on other aspects of their projects.
Sample Processing QC
Sample processing QC assesses various attributes of a sample to gauge its overall quality, tailored to sample types and client needs. Quality control is performed before and after several steps in the workflow, ensuring high sample quality. Sampled conducts stringent multifactorial quality control checks, ensuring accurate and reliable data for clients.
For DNA QC, we utilize the Lunatic automated UV spectrometer and an automated BioMicroLab non-contact volume detection instrument to assess concentration and volume, complemented by functional quality control for accurate DNA quality evaluation. RNA QC follows a similar process, with the Lunatic spectrometer, volume assessment, and RNA quality score via the Perkin Elmer LabChip GX for RNA fragmentation. Plasma and serum QC includes visual grading and analytical quality control for bilirubin, hemoglobin, and turbidity. Results are accessible via SampledSphere, allowing clients to decide on aliquoting, storage, shipping, or direct laboratory analysis based on their project needs.
Aliquoting
Aliquoting involves dividing a sample into several separate tubes for storage or analysis, safeguarding against processing risks like freeze-thaw cycles. It plays a central role in project management, allowing sample preservation for future use. Sampled’s high-capacity automated aliquoting enhances efficiency and maintains sample quality for versatile applications.
In our aliquoting workflow, we handle samples received from various sources, ensuring proper storage throughout the handling process. Using the Hamilton instrument, we offer clients the flexibility to create precise aliquots at varying volumes and container types, optimizing accuracy and throughput. Manual aliquoting is also available when it aligns with specific client needs. After aliquoting, samples can be directed for analysis, storage, or shipping to the client or a third party, expertly coordinated by our logistics team.
