What Is iPSC Reprogramming?
Induced pluripotent stem cell (iPSC) reprogramming is the process where somatic cells are reprogrammed to a stem cell or pluripotent state.
iPSC generation can be performed using chemical or genetic methods and is used for a variety of applications in academia, industry, and the clinic. The generation of iPSCs is a time-consuming and technically challenging process requiring high levels of experience and technical expertise.
At Sampled, we can derive iPSCs from any cell type, but our standard workflow is with whole blood and skin biopsies. We perform robust quality control (QC) on clones to ensure the best quality iPSC lines are delivered. Sampled guarantees the generation of suitable clones that meet our clients’ requirements. Our clients typically use iPSCs for disease and development modeling, but our service supports a broad range of applications.

In addition, through our U24 cooperative agreement with NINDS, we distribute a GMP cell line that can be used for clinical trials. The cell line was generated by Lonza for NINDS and is distributed by Sampled.

How Does iPSC Reprogramming Work?
Client Parameters
The iPSC service at Sampled begins with defining the requirements of our client. This includes determining parameters such as:
- Source material – what cell type is being used and in what format (i.e., skin biopsy or isolated fibroblasts)?
- Number of clones – we offer a 1 or 3 clone service.
- Quality control – all iPSCs generated come with our standard QC, plus additional QC based on client needs/requests.
- Scale – we offer small (6-8 vials) and large (40-60 vials) scale.
Source Material
At Sampled we can work with any cell type and typically work with skin biopsies and whole blood. However, we are happy to receive cells or material from any step of the iPSC pipeline. This includes but is not limited to peripheral blood mononuclear cells (PBMCs), lymphoblastoid cell lines (LCLs), and fibroblasts isolated by our clients or a third party.
Quality Control
We offer wide-ranging QC options for our clients. Our standard QC protocol includes:
- Sterility testing
- Mycoplasma testing
- Viability
- FACS for Stem cell marker
- Alkaline phosphatase staining
- Identity (via SNPTrace)
Additional optional QC we offer include:
- Karyotyping
- Embryoid body formation (scorecard for germ layer differentiation)
- Pluritest array
- Sendai persistence (for blood-derived iPSCs)
- Episome persistence (for fibroblast-derived iPSCs)
- CNV array
Timescale
The timescale for iPSC generation depends on the client’s requirements. The timescale for a single clone with standard QC is 3-4 months. 3 clones with full QC can take up to 6 months. Our systems are optimized for iPSCs derived from blood and skin and therefore other sources may require additional optimization steps and take longer to perform.
