iPSC Reprograming

Sampled’s team of experts is ready to reprogram any cell type into induced pluripotent stem cells (iPSCs) for use in academic and industry research environments. At Sampled, we perform robust quality control on our iPSCs and guarantee the generation of suitable clones.

What Is iPSC Reprogramming?

Induced pluripotent stem cell (iPSC) reprogramming is the process where somatic cells are reprogrammed to a stem cell or pluripotent state.

iPSC generation can be performed using chemical or genetic methods and is used for a variety of applications in academia, industry, and the clinic. The generation of iPSCs is a time-consuming and technically challenging process requiring high levels of experience and technical expertise.

At Sampled, we can derive iPSCs from any cell type, but our standard workflow is with whole blood and skin biopsies. We perform robust quality control (QC) on clones to ensure the best quality iPSC lines are delivered. Sampled guarantees the generation of suitable clones that meet our clients’ requirements. Our clients typically use iPSCs for disease and development modeling, but our service supports a broad range of applications.

Why Use Sampled’s iPSC Reprogramming Service?

Expertise

Sampled’s team of experts can generate iPSCs from any cell type. Clients trust us with difficult-to-transform cell lines and samples with low cell counts. At Sampled, we achieve consistently high success rates for iPSC generation and have the experience to efficiently troubleshoot issues as they arise.

Time- and Cost-efficient

iPSC generation is a technically difficult and a time-consuming process. It is also a costly platform to develop in-house in terms of the money and time taken to train technicians and troubleshoot issues. By outsourcing your iPSC generation needs to Sampled, you can use your time and resources to focus on other things while ensuring your sample is in the hands of global experts using state-of-the-art processes.

Quality Control

At Sampled, all our iPSCs come with our standard QC. Additionally, we offer a stringent QC process for our clients depending on their needs. Good QC is essential for the success of future experiments.

Guaranteed

At Sampled, we guarantee that our iPSC reprogramming service will provide clients with their desired clones. Our team of experts will continue to screen clones until we have the expected clones.

Trusted

In addition to our hundreds of clients, Sample is also trusted as the sole repository for the National Institute of Neurological Disorders and Stroke (NINDS) and the National Institute of Mental Health (NIMH).

iPSC Reprogramming Applications

The applications for our iPSC reprogramming service are research-only and we work with academic and industry laboratories.

In addition, through our U24 cooperative agreement with NINDS, we distribute a GMP cell line that can be used for clinical trials. The cell line was generated by Lonza for NINDS and is distributed by Sampled.

Applications for our services include:

  • Clinical trials
  • Research studies
  • Biomarker discovery
  • Population health studies
  • Agrigenomics
  • Disease modeling
  • Development modeling

How Does iPSC Reprogramming Work?

Client Parameters

The iPSC service at Sampled begins with defining the requirements of our client. This includes determining parameters such as:

  • Source material – what cell type is being used and in what format (i.e., skin biopsy or isolated fibroblasts)?
  • Number of clones – we offer a 1 or 3 clone service.
  • Quality control –  all iPSCs generated come with our standard QC, plus additional QC based on client needs/requests.
  • Scale – we offer small (6-8 vials) and large (40-60 vials) scale.

Source Material

At Sampled we can work with any cell type and typically work with skin biopsies and whole blood. However, we are happy to receive cells or material from any step of the iPSC pipeline. This includes but is not limited to peripheral blood mononuclear cells (PBMCs), lymphoblastoid cell lines (LCLs), and fibroblasts isolated by our clients or a third party.

Quality Control

We offer wide-ranging QC options for our clients. Our standard QC protocol includes:

  • Sterility testing
  • Mycoplasma testing
  • Viability
  • FACS for Stem cell marker
  • Alkaline phosphatase staining
  • Identity (via SNPTrace)

Additional optional QC we offer include:

  • Karyotyping
  • Embryoid body formation (scorecard for germ layer differentiation)
  • Pluritest array
  • Sendai persistence (for blood-derived iPSCs)
  • Episome persistence (for fibroblast-derived iPSCs)
  • CNV array

Timescale

The timescale for iPSC generation depends on the client’s requirements. The timescale for a single clone with standard QC is 3-4 months. 3 clones with full QC can take up to 6 months. Our systems are optimized for iPSCs derived from blood and skin and therefore other sources may require additional optimization steps and take longer to perform.
