What is Blood Fractionation?

Fractionation involves the centrifugation of whole blood into separate fractions, usually to acquire serum or plasma.

For serum, a silicone gel within the centrifugation tube is typically used to keep the serum separated from the rest of the sample upon centrifugation. Unclotted blood separates into plasma, buffy coat (containing white blood cells), and red blood cells upon centrifugation without using a silicone gel.

Serum and plasma are both light-yellow in color but differ in their composition and suitability for downstream applications. Serum is the liquid that remains when clotted blood is centrifuged and plasma is the liquid that remains when unclotted blood is centrifuged as mentioned above.

Sampled can perform quality control on serum and plasma depending on our client’s requirements. We perform visual color grading of plasma, and analytical control using a Lunatic instrument from Unchained Labs which tests levels of bilirubin and hemoglobin, as well as the turbidity of the sample.

How Does Fractionation Work?

Sample Receipt

All samples we receive must arrive at room temperature. If they arrive frozen, subsequent thawing causes hemolysis which makes fractionation impossible. We also recommend sending us whole blood instead of plasma since frozen plasma is highly susceptible to degradation upon thawing. Upon receipt, the whole blood sample is maintained at room temperature and processed within 48 hours.

Fractionation

Whole blood fractionation is performed by an automated instrument operated by one of Sampled’s technical experts. Our Hamilton easyBlood STARlet™ liquid handling platforms recognize the different layers of fractionated blood which enables it to safely aliquot the serum or plasma. Sampled’s technical expertise and high-end instrumentation means your samples are processed quickly and safely.

Further Processing

After fractionation, the sample can be sent immediately for analysis, storage, or shipping to the client or a third party chosen by the client. Fractionation of whole blood is an essential prerequisite for analyzing cell-free DNA.
