PCR
Sampled excels in PCR services, offering cutting-edge droplet digital PCR (ddPCR) for ultra-sensitive analysis and high-end quantitative PCR (qPCR) using Applied Biosystems technology for precise gene-expression studies. Our expert team ensures accurate, reliable results for diverse research needs.
What Are Sampled’s PCR Services?
Sampled’s PCR services cover qPCR and ddPCR, which give our clients crucial insights into their samples, including gene expression analyses and detection of structural variants.
qPCR
qPCR is a highly sensitive technique used to detect specific DNA sequences in a sample. It enables gene expression analysis, and it can also determine absolute target DNA abundance. Offering advantages over conventional PCR, qPCR provides real-time insights into DNA levels. Sampled employs qPCR as a versatile and powerful tool in our multi-omics arsenal, adhering to global standards such as the MIQE guidelines for accurate and reliable results in gene validation and expression studies.
At Sampled, we maintain robust handling and processing standards for DNA and RNA, in addition to rigorous quality control, to ensure we use only high-quality input material for qPCR analysis. If we receive RNA or raw samples, we perform reverse transcription and normalize DNA concentration to achieve optimal amplification detection. Our qPCR platform offers flexibility with chemistries, and by accommodating 96- and 384-well formats for high throughput analyses. Post-qPCR, clients receive raw results or expert-performed analyses, meeting the highest quality standards.
Droplet Digital PCR
ddPCR is an advanced technology for ultra-sensitive nucleic acid detection and absolute quantification. Through precise partitioning of samples into individual droplets, ddPCR achieves unparalleled sensitivity, making it ideal for detecting low-abundance targets like allelic variants. Its ability to perform absolute quantification sets it apart and is crucial in applications requiring accurate target molecule counts. Sampled’s expert team is well-equipped to support diverse ddPCR projects, ensuring precision and reliability in genetic research, clinical diagnosis, and environmental science.
Sampled excels in ddPCR, handling diverse samples with in-house or client-provided DNA, subject to robust quality control. Sample and droplet generation involves meticulous preparation and loading into the QX200 Droplet Generator, creating 20,000 uniform droplets. PCR amplifies these droplets in a 96-well plate, which is followed by droplet reading on the QX200 Droplet Reader, allowing simultaneous examination of multiple targets. Expert bioinformaticians at Sampled analyze data, plotting fluorescence-based positive and negative droplets, providing clients with comprehensive and statistically sound insights.

What Are the Input Requirements for PCR?
Item | Type |
---|---|
Sample Types | High-quality DNA sourced from blood, serum, saliva, tissue biopsies, hair, nail, stool, urine, semen, and cell line samples/pellets, buccal swabs, and environmental samples. We also work with pre-extracted DNA. |
Storage/Transport Conditions | Pre-extracted DNA should be transported at 4°C (or as close to the previous storage conditions as possible). RNA should be stored at -20°C and transported on dry ice. For information on other tissue types, please contact a member of our team. |
Input Requirements | Input depends on the application and abundance of target DNA. |
